  3. Our research is now supported by a 2024 Simons Foundation Autism Research Initiative (SFARI) Pilot Award, co-funded by the Nancy Lurie Marks Family Foundation (NLMFF). I am grateful to both SFARI and NLMFF for investing in high-risk, high-reward science to advance understanding of autism spectrum disorders (ASD) and neurodevelopmental disorders.

    The project will explore how mutations in chromatin regulators ASH1L and SETBP1 impair their crucial role in guiding the development of GABAergic neurons—key cells maintaining the brain’s balance between excitation and inhibition. Disruptions in this process are strongly linked to ASD, but the molecular pathways remain poorly understood.

    By integrating genomic, proteomic, and functional approaches, we aim to reveal how these enzymes regulate gene activity during inhibitory neuron development and how their dysfunction contributes to ASD.

    We discovered that #marijuana (THC specifically) reduces #GABA #neuron activity. This helps explain some of the rewarding and calming effects of THC because the depression of GABA #neurons means increased dopamine release, which generally makes us feel pretty good.

    Don't smoke too young though, we also found #THC is MUCH more influential in adolescents, and sometimes struggles to reverse!

  12. Yesterday I tried #GABA Shen puehr, and what I felt was the milder version of the Simao #Shen #puehr #tea I already have. I didn’t feel the bright sweetness which I usually notice in a well-fermented GABA. I have one more portion of the tea, and maybe I’ll feel more the next time.😊

  15. In response to a few questions people have asked:

    We used whole-cell patch-clamp #electrophysiology to record the activity of single #neurons.

    The "resist changes" refers to #neuroplasticity. We found a reduction in long-term depression, or #LTD, in adult mice after high frequency stimulus (HFS) compared to adolescent mice.

    These cells are specifically #GABA releasing cells in the ventral tegmental area (#VTA ), where GABA can regulate dopamine release throughout the #brain.

  16. @P_KA_T It is related in the sense that while frequently used pathways tend to fire more easily, there are controls on those pathways from regulatory inhibition, which is what we studied. So even if a synapse may exhibit relatively high activity from frequent use, they can be told to "slow down' by GABA, which we determined was less adaptive in adults.

    Also, should we not just cave and call them #inhibitory cells rather than #interneurons, given the importance of #gaba ergic projection neurons? (Not trolling the developmental people who see gaba excitation).
