In a mouse experiment where obesity was induced with a high-fat diet, subsequent addition of epigallocatechin-3-gallate (EGCG)--a compound found in green tea--to the diet for four weeks resulted in lower body weight, better insulin sensitivity, no hippocampal shrinkage, and better memory. The brains of the EGCG-treated mice had lower inflammation and normal cellular recycling.

  6. #Wegovy®, #Zepbound®, and similar medications continue to gain attention for effectively treating #obesity, and now there is discussion—with research underway—about their potential for improving certain #surgical outcomes.

    The brand-name medications mentioned above belong to a class called GLP-1 (glucagon-like peptide-1) agonists. #GLP-1s mimic the actions of the GLP-1 hormone, which is secreted in the gut and sends an “I’m full” signal to the brain after you eat.

    GLP-1 medications aren’t new. @Ozempic® (generic name: #semaglutide) received approval from the U.S. Food and Drug Administration (FDA) to treat type 2 diabetes in 2017.

    After its weight-loss effects were studied, semaglutide (at a higher dose and sold under the name Wegovy) was approved by the FDA as an anti-obesity medication in 2021, and others have since followed.

    Most recently, the FDA approved Wegovy for patients with #cardiovascular #disease and obesity or overweight after a study showed that those taking the medication had a lower overall risk of major cardiac events, such as heart attack, stroke, or cardiovascular-related death.

    Now, some Yale Medicine surgeons believe, based on their ongoing research, that taking anti-obesity medications before and sometimes after total #joint #replacement and #bariatric #surgery can be beneficial.

    “These medications have been transformative, in terms of expanding the pool of people who can safely undergo joint replacement surgery,” says Daniel Wiznia, MD, a Yale Medicine orthopaedic surgeon and co-director of the Avascular Necrosis Program.

    “These anti-obesity medications are game-changers; they are changing the way people practice medicine.”

    Often, patients who need a total hip or knee replacement must delay surgery until they lose enough weight to meet specific body mass index ( #BMI ) standards.
    This is important in protecting the patient’s health.

    “Weight increases a patient’s risk of postoperative complications, including wound-healing problems, stroke, heart attack, blood clots, and infection,” he says.

    However, having patients take a GLP-1 medication before surgery, as well as improving their diet, has allowed more patients to reach a safe preoperative BMI, Dr. Wiznia says.

Sahu et al. now report that pyrimidines influence the activity of pyruvate dehydrogenase, a key intermediary enzyme between glycolysis and the tricarboxylic acid cycle and respiration. Depletion of pyrimidines led to loss of thiamine pyrophosphate (a required cofactor for pyruvate dehydrogenase), apparently because the pyrimidine UTP rather than ATP functions as a preferred substrate for TPK1, the enzyme that produces thiamine pyrophosphate. ..."

    Some ppl report improvement in chronic health problems with thiamine or d-ribose supplementation. I've always been wary of the latter due to concerns about cancer-promotion and also neurological harms, as D-ribose increases production of advanced glycation end products.

    Thiamine seems to have a biphasic dose response with respect to cancer-promotion, at least in vitro.

    Between 2020 and 2022, the world’s most affluent 1% of people captured nearly twice as much of the new global wealth created as did the other 99% of individuals put together
    And in 2019 they emitted as much #carbon #dioxide as the poorest two-thirds of humanity.
    In the decade to 2022, the world’s billionaires more than doubled their wealth, to almost US$12 trillion.

    The evidence gathered by social epidemiologists, including us, shows that large differences in income are a powerful #social #stressor that is increasingly rendering societies dysfunctional.
    For example, bigger gaps between rich and poor are accompanied by higher rates of #homicide and #imprisonment.
    They also correspond to more #infant #mortality, #obesity, #drug #abuse and @COVID-19 #deaths, as well as higher rates of teenage pregnancy and lower levels of child well-being, social mobility and public trust
    The #homicide rate in the United States
    — the most unequal Western democracy
    — is more than 11 times that in Norway (see go.nature.com/49fuujr).
    #Imprisonment rates are ten times as high, and infant mortality and obesity rates twice as high.

    These problems don’t just hit the poorest individuals, although the poorest are most badly affected.
    Even #affluent people would enjoy a better quality of life if they lived in a country with a more equal distribution of wealth, similar to a Scandinavian nation.
    They might see improvements in their mental health and have a reduced chance of becoming victims of violence; their children might do better at school and be less likely to take dangerous drugs.

    The costs of inequality are also excruciatingly high for #governments.
    For example, the Equality Trust, a charity based in London (of which we are patrons and co-founders), estimated that the United Kingdom alone could save more than £100 billion ($126 billion) per year if it reduced its inequalities to the average of those in the five countries in the Organisation for Economic Co-operation and Development (OECD) that have the smallest income differentials
    — Denmark, Finland, Belgium, Norway and the Netherlands5.
    And that is considering just four areas: greater number of years lived in full health, better mental health, reduced homicide rates and lower imprisonment rates.
    Many commentators have drawn attention to the environmental need to limit economic growth and instead prioritize sustainability and well-being.
    Here we argue that tackling inequality is the foremost task of that transformation.
    Greater @equality will reduce unhealthy and excess consumption, and will increase the #solidarity and #cohesion that are needed to make societies more adaptable in the face of climate and other emergencies.
