“I didn’t know she was Black until a number of years ago 🔸when she happened to turn Black🔸 and now she wants to be known as Black.
So I don’t know, is she Indian or is she Black?” Mr. Trump said of Ms. Harris, whose mother was Indian American and whose father is Black. The moment was shocking, but for those who have followed Mr. Trump’s divisive language,
it was hardly surprising.♦️The former president has a history of using race to pit groups of Americans against one another, amplifying a strain of racial politics that has risen as a generation of Black politicians has ascended♦️
The audacity of Mr. Trump, a white man, questioning how much a Black woman truly belongs to Black America was particularly incendiary.
And it evoked 🔷an ugly history in this country, in which white America has often declared the racial categories that define citizens, and sought to determine who gets to call themselves what.🔷
“Give me a break,” said Fred Sweets, a contributing editor at The St. Louis American who watched the discussion from the third row.
“He seemed to be denigrating her background. She knows who she is.”Ms. Harris has embraced her dual racial identities.
She has long identified as Black and was shaped by several Black institutions. She graduated from Howard University, a historically Black university in Washington, D.C., and there joined Alpha Kappa Alpha, the nation’s oldest Black sorority.
She has spoken extensively about growing up in what she described as a Black community in Berkeley, Calif.
"She had two Black babies, and she raised them to be two Black women,” Harris told The New York Times in a 2016 interview about her mother.
On Wednesday evening, Ms. Harris responded to Mr. Trump’s comment at an event hosted by one of the nation’s most prominent Black sororities, saying they showed “#divisiveness and #disrespect.”
“The American people deserve a leader who tells the truth,” she said, making no direct reference to Mr. Trump’s personal attacks.
“We deserve a leader who understands that our differences do not divide us
— they are an essential source of our strength.”Attacks on Ms. Harris’s racial background have circulated among right-wing figures and Mr. Trump’s close allies for years.
In 2019, Donald Trump Jr. shared a social media post from an alt-right personality that falsely claimed Ms. Harris was
🔥not Black enough 🔥to be discussing the plight of Black Americans during a primary debate.Though Mr. Trump later deleted the post, it spread widely across conservative social media, prompting a wave of accounts to question her background,
👉which was exactly the point of the effort, according to some far-right activists.

CAN YOU NAME AN ANIME STARTING WITH THE LETTER K?
I’ll start with #KillLaKillThere were a lot of different anime mentioned for last week's letter C and I am now looking forward to this week's letter K. I wonder how many anime there are starting with a K.

👋DefenseFinder news !
Thinking of how we can better help the community, we provide a new website that includes a wiki on defense systems, a structure database and precomputed DefenseFinder results on >22k genomes.
https://defensefinder.mdmlab.fr
Here is the preprint : https://www.biorxiv.org/content/10.1101/2024.01.25.577194v1
In the ever evolving antiphage defense landscape, we’ve added 92 new systems, now detecting a total of 152. These additions account for more than 33% of the systems detected. Please keep telling us if we are missing some. Full list here: https://defensefinder.mdmlab.fr/wiki/defense-systems
The new web service interface allows anyone to detect defense systems in their preferred genomes and to save previous analysis (without any registration). We also provide genomic visualization and the ability to easily download everything.
One major feedback we had, was the difficulty to understand the hits identified by DefenseFinder and put it in context.
We created a wiki (https://defensefinder.mdmlab.fr/wiki/) where each defense system has its page. Systems found by DefenseFinder are now linked to those wiki-like pages.
The Wiki is usable on its own, allowing research through the diversity of system mechanisms and domains through a searchable table. We added general concepts often used in the field.
It’s a collaborative and participative website, so create missing pages or tell us about them ✍️!
You can find, on the help page of the website, how to contribute. Don’t hesitate to ask us in case of problem.
To allow direct search of the presence of systems in public genomes, we ran DefenseFinder on RefSeq complete genomes database. We provide visualization through interactive tables and graphs that are easily downloadable.
We now live in a world of structure predictions, so we teamed up with Gemma Atkinson’s lab and precomputed all monomers as well as dimers for all known representatives of defense systems. Foldseek results on the AlphaFoldDB and PDB were also precomputed. All these results can be visualized with Mol*, a powerful tool-kit for web-based visualization of protein structures.
Congrats to Florian Tesson, lead author and Rémi Planel for the website development as well as the IT Department of the Institut Pasteur for the technical part.
We organized a hackathon day, internal to the lab, to fill the 152 defense systems’ pages. We also ask expert in the field to write the page on their favorite system. So : huge thanks to everyone who participated in the project, of course MDM Lab members but also wiki contributors : Adi Millman, François Rousset, Daan C. Swarts, Avigail Stokar-Avihail, Nathalie Béchon, Alex L. Gao
We really hope this web service will allow everyone to dive into antiphage defense biology by having easy access to bioinformatics resources.
Don’t hesitate to let us know what you would like to see next and join the fun of creating improving wiki pages!

Serious diseases—ranging from emphysema and cystic fibrosis to Alzheimer's disease—can result when the 🔹cellular oversight of protein folding goes awry🔹.
Identifying the glyco-code responsible for high-fidelity folding and quality control could be a promising way for drug therapies to target many diseases.
Scientists once thought that the single code governing life was #DNA, and that everything was governed by how DNA's four building blocks—A, C, G and T—combined and recombined.
But in recent decades, it has become clear that there are other codes at work, and especially in building the intricately folded, secreted proteins that are created in 🔸the human cell's protein factory, the "endoplasmic reticulum" (ER)🔸, a membrane-enclosed compartment where protein folding begins.
Approximately 7,000 different proteins—one-third of all the proteins in the human body—mature in the ER.
The secreted proteins—collectively known as the "#secretome"—are responsible for everything from our body's enzymes to its immune and digestive systems and must be formed correctly for the human body to function normally.Special molecules called "#chaperones," help fold the protein into its final shape.
They also help to identify proteins that haven't folded quite correctly, lending them additional help in refolding, or, if they're hopelessly misfolded, targeting them for destruction before they cause damage.
However, the chaperone system itself, which comprises a part of the cell's quality control department, sometimes fails, and when it does, the results can be catastrophic for our health.
The discovery of the carbohydrate-based chaperone system in the ER was due to the pioneering work that 👉Daniel Hebert, professor of biochemistry and molecular biology at UMass Amherst and one of the paper's senior authors, initiated as a postdoctoral fellow in the 1990s.
"The tools we have now, including glycoproteomics and mass spectrometry at UMass Amherst's Institute for Applied Life Sciences, are allowing us to answer questions that have remained open for over 25 years," says Hebert. "The lead author of this new paper, 👉Kevin Guay, is doing things I could only dream of when I first started."
Among the most pressing of these unanswered questions is: ♦️how do chaperones know when 7,000 different origami-like proteins are correctly folded?♦️
We know now that the answer involves an "ER gatekeeper" enzyme known as #UGGT, and a host of carbohydrate tags, called "N-glycans", which are linked to specific sites in the protein's amino acid sequence.Guay, who is completing his Ph.D. in the molecular cellular biology program at UMass Amherst, focused on two specific mammalian proteins, known as "alpha-1 antitrypsin" and "antithrombin".
⭐️Using CRISPR-edited cells, he and his co-authors modified the ER chaperone network to determine how the presence and location of N-glycans affected protein folding.
They watched as the disease variants were recognized by the ER gatekeeper UGGT and, in order to peer more closely, developed a number of innovative glycoproteomics techniques using mass spectrometry to understand what happens to the glycans that stud the surface of the proteins.
What they discovered is that ⚠️the enzyme UGGT "tags" misfolded proteins with sugars placed in specific positions. ⚠️
It's a sort of code that the chaperones can then read to determine exactly where the folding process went wrong and how to fix it.
"This is the first time that we've been able to see where UGGT puts sugars on proteins made in human cells for quality control," says Guay.
"We now have a platform for extending our understanding of how sugar tags can send proteins for further quality control steps and our work suggests that UGGT is a promising avenue for targeted drug therapy research."
"What's so exciting about this research," says👉 Lila Gierasch, distinguished professor of biochemistry and molecular biology at UMass Amherst and one of the paper's co-authors, "is the discovery that glycans act as a code for protein folding in the ER.
The discovery of the role that UGGT plays opens the door to future advancement in understanding and eventually treating the hundreds of diseases that result from misfolded proteins."
