#Age-Dependent #Pathogenesis of #Influenza A Virus #H7N9 Mediated Through #PB1-F2-Induced Mitochondrial DNA Release and Activation of cGAS-STING-NF-κB Signaling
Source: Journal of Medical Virology, ABSTRACTExactly why human infection of avian influenza A virus H7N9 causes more severe disease in the elderly remains elusive. In this study, we found that H7N9 PB1-F2 is a pathogenic factor in 15–18-month-old BALB/C mice (aged mice) but not in 6–8-week-old young adult mice…

Enhanced Diversifying #Selection on #Polymerase Genes in #H5N1 Clade 2.3.4.4b: A Key Driver of Altered #Species #Tropism and #Host Range Expansion, BioRxIV: https://www.biorxiv.org/content/10.1101/2024.08.19.606826v1?rss=1
the polymerase genes #PB2, #PB1, and #PA exhibit significantly greater selection pressures in clade 2.3.4.4b than in all the earlier H5N1 virus clades. Polymerases play critical roles in influenza virus adaptation, including viral fitness, interspecies transmission, and virulence.

High #Prevalence of #HPAI: A Virus in #Vietnam's Live #Bird #Markets, Open Forum Infect Dis.: https://academic.oup.com/ofid/article/11/7/ofae355/7700791?login=false
...a positive #human #sample had a #H9N2 #avian-like #PB1 #gene. In contrast, #prevalence of influenza A in the #swine #farms was much lower with only 6 (0.4%) of the 1700 total swine farm samples studied, having molecular evidence for influenza A virus.

#Aminoacid #mutations #PB1-V719M and #PA-N444D combined with #PB2-627K contribute to the #pathogenicity of #H7N9 in #mice, Vet Res.: https://veterinaryresearch.biomedcentral.com/articles/10.1186/s13567-024-01342-6
Overall, this study revealed that #virulence in H7N9 is a #polygenic trait and identified novel virulence-related residues (PB2-627K combined with PB1-719M and/or PA-444D) in viral ribonucleoprotein (vRNP) complexes.

Identification of a novel #reassortant #H3N2 avian #influenza virus based on nanopore sequencing technology & genetic characterization, Zhonghua Liu Xing Bing Xue Za Zhi, https://pubmed.ncbi.nlm.nih.gov/38678355/
This study identified a novel reassortant H3N2 avian influenza virus by nanopore sequencing, with #PB1 gene derived from #H5N6 avian influenza virus. Virus had a low ability to spread across species, but further exploration was needed to determine whether its pathogenicity to the host was affected.
