  14. Within tumors in the human body, there are #immune #cells ( #macrophages ) capable of fighting cancer,
but they have been unable to perform their roles properly due to suppression by the tumor.

KAIST researchers have overcome this limitation by developing a new therapeutic approach that directly ⚠️converts immune cells inside tumors into anticancer cell therapies.

KAIST President Kwang Hyung Lee announced on the 30th that a research team led by Professor Ji-Ho Park of the Department of Bio and Brain Engineering has developed a therapy in which,
when a drug is injected directly into a tumor, macrophages already present in the body absorb it, produce CAR
(a #cancer-#recognizing device) proteins on their own...
and are converted into anticancer immune cells known as "#CAR-#macrophages."

❌Solid tumors—such as gastric, lung, and liver cancers—grow as dense masses, making it difficult for immune cells to infiltrate tumors or maintain their function.

As a result, the effectiveness of existing immune cell therapies has been limited.

    ✅ CAR-macrophages,
which have recently attracted attention as a next-generation immunotherapy,
have the advantage of directly engulfing cancer cells while simultaneously activating surrounding immune cells to amplify anticancer responses.

⛔️However, conventional CAR-macrophage therapies require immune cells to be extracted from a patient's blood, followed by cell culture and genetic modification.

This process is time-consuming, costly, and has limited feasibility for
    real-world patient applications.

To address this challenge, the research team focused on "#tumor-#associated #macrophages" that are already accumulated around tumors.

⭐️They developed a strategy to directly reprogram immune cells in the body by loading lipid nanoparticles
—designed to be readily absorbed by macrophages
    —with both mRNA encoding
    cancer-recognition information
and an immunostimulant that activates immune responses.

In other words, in this study,
    CAR-macrophages were created by
👉"directly converting the body's own macrophages into anticancer cell therapies inside the body."

  22. 2025-8-9 Is this MCAS anaphylaxis, anxiety, or anaphylaxis from anxiety?

I almost didn’t want to share this one, because I’m in a bit of a nervous state, and I was worried about the people who think people with Long Covid are psychological malingerers, etc. When I had to leave work, and filed for US Social Security Disability, I kept my Generalized Anxiety Disorder out of my initial application, afraid of getting the dreaded “just anxiety,” dismissal that many complex chronic illness kin are familiar with. (I was later told this was a bad move, and to definitely include all mental health and neurodivergence things, which I did for the ongoing process.)

Even as I’m having less anaphylaxis risk, it’s taking time to recover psychologically. I told my therapist how I felt like I backslid over a decade in mental health work overnight from the first recent anaphylaxis episode. She kindly said that while I might feel as scared and threatened as I often did back then, that the trigger was different. I was not feeling that anxious about an email, I was feeling that way about a literal life and death situation, one which I still don’t fully understand the triggers of.

A decade ago, de-stigmatizing mental healthcare was really important to me too. Now, I’m especially aware of how much these issues intersect, so heck yeah, publishing the medical art about anxiety.

Lastly, I wanted to include few examples of the ways in which these issues intersect. These are absolutely the tip of the iceberg.

    • Postural Orthostatic Tachycardia Syndrome, or POTS, is a form of autonomic dysfuntction (when things your body normally does automatically stop behaving normally), and is a part of my Long Covid. It involves a high heart rate, and often symptoms like room spinning or lightheadedness, trembling or shaking, shortness of breath, and poor bloodflow. POTS is often misdiagnosed as anxiety, and The Disabled Ginger wrote about it here.

Theories that diseases are caused by mental states and can be cured by will power are always an index of how much is not understood about the physical terrain of a disease.

Moreover, there is a peculiarly modern predilection for psychological explanations of a disease, as of everything else. Psychologizing seems to provide control over the experiences and events (like grave illnesses) over which people have in fact little or no control. Psychological understanding undermines the ‘reality’ of a disease. That reality has to be explained.

Susan Sontag, illness as metaphor (New York: Farrar, Straus and Giroux, 1977), p. 55.

p.s. mental health is physical health and physical health is mental health. you got a literal brain in your body and all of your body is involved in everything that you do. please take care of yourselves and each other. <3

  28. Still reeling from #PFAS spill, #BrunswickMaine residents push back on plans for #sludge treatment plant

by AnnMarie Hilton, Tue, January 28, 2025

"Brunswick resident Sandra Carslick said she and her neighbors have been living in a 'bad dream' for the five months since 1,600 gallons of toxic foam spilled so-called forever chemicals at the already contaminated former Naval Air Station nearby.

"At a Brunswick Town Council meeting on Monday night, Carslick said that bad dream could soon become residents’ 'worst nightmare,' if the town reopens a processing site for chemical-laden [#Biosolids] sludge from sewer treatment plants across Maine, and potentially beyond.

"The residents were responding to plans from Delaware-based #ViridiEnergy to update and expand #BrunswickLanding’s anaerobic digester, which converts sewage sludge — also referred to as #biosolids — into renewable natural gas and byproducts that get sent to landfills.

"During Monday’s presentation to the town council, company representatives explained how the facility would work and what local residents could expect. Several councilors said they would like to see additional data before deciding, but some already expressed concerns that the project isn’t right for Brunswick, particularly at this time. Members of the public also spoke for more than an hour, overwhelmingly concerned about the project and its potential to bring more #HarmfulChemicals into the area.
"The town is still dealing with the accidental discharge last summer of #firefighting foam containing #perfluoroalkyl and #polyfluoroalkyl substances, also known as #PFAS, at a hangar at #BrunswickExecutiveAirport, which is also situated at Brunswick Landing.

"The spill has prompted environmental and public health concerns over the PFAS, which have been linked to serious long-term health problems including #cancer, weakened #immune systems, developmental issues, and more. Since it started surveying and reviewing the material in the late 1980s, the U.S. Environmental Protection Agency [#EPA] has found more than 700 chemicals in sewage sludge, including PFAS.

"Chet Benham, a senior advisor and one of the founders of Viridi, tried to address those concerns during the meeting, telling residents the facility will operate according to all state and local requirements. He said there are protocols to prevent spills of the sludge that will move through the closed-loop facility.

"'We feel really, really good about it,' Benham said. 'We wouldn’t be standing here before you if we didn’t.' [Typical #Greenwashing #Bullshit]

"According to Viridi’s plan, the facility would take in about 85,000 tons of biosolids per year, which would be processed into renewable natural gas that would feed into Maine Natural Gas. About 10,000 tons of solid material byproduct would be sent from the plant to the #JuniperRidgeLandfill in Old Town. Over the course of the discussion, it came to light that Viridi would also likely bring in sludge from outside of Maine in order to run their facility at full capacity."

  30. “It’s difficult to think of a disease, injury or injection that doesn’t involve some kind of #immune #response,
and our finding really changes the way we could control this response,” said Professor Adrian Liston from the University’s Department of Pathology and the study’s corresponding author.

“We’ve uncovered new rules of the immune system.
This ‘unified healer army’ can do everything
    – repair injured muscle, make your fat cells respond better to insulin, regrow hair follicles.

To think that we could use it in such an enormous range of diseases is fantastic:
it’s got the potential to be used for almost everything.”

Lymphoid organs are integral parts of the immune system, responsible for producing #lymphocytes, a type of white blood cell that includes T cells.

T cells begin life in the bone marrow and then move to the thymus, an organ in the upper mid-chest,
where they mature into specialized subsets, including #Tregs.

Once fully matured, T cells are exported to peripheral lymphatic tissues and organs like the spleen, tonsils, and lymph nodes
    (some move to the bloodstream).

It was thought that’s where Tregs stayed, on ‘standby’ until called upon by the immune system.

To test this, the researchers analyzed the Tregs present in 48 different tissues in mice, including lymphoid and non-lymphoid tissues and tissues associated with the gut.

They found them in all tissue types, suggesting that Tregs weren’t specialized cell populations confined to lymphoid tissues but moved around the body, executing repair functions in areas that need it.

“Now that we know these regulatory T cells are present everywhere in the body, in principle ⭐️we can start to make immune suppression and tissue regeneration treatments that are targeted against a single organ⭐️
    – a vast improvement on current treatments that are like hitting the body with a sledgehammer,” Liston said

  32. #Cannabinoids as novel #AntiInflammatory drugs

    by Prakash Nagarkatti, et al.
    October, 2009

    Abstract:

"Cannabinoids are a group of compounds that mediate their effects through cannabinoid receptors. The discovery of Δ9-tetrahydrocannabinol (#THC) as the major psychoactive principle in marijuana, as well as the identification of #cannabinoid receptors and their endogenous ligands, has led to a significant growth in research aimed at understanding the physiological functions of cannabinoids. Cannabinoid receptors include CB1, which is predominantly expressed in the brain, and CB2, which is primarily found on the cells of the immune system. The fact that both CB1 and CB2 receptors have been found on #immune cells suggests that cannabinoids play an important role in the regulation of the immune system. Recent studies demonstrated that administration of THC into mice triggered marked apoptosis in T cells and dendritic cells, resulting in immunosuppression. In addition, several studies showed that cannabinoids downregulate #cytokine and chemokine production and, in some models, upregulate T-regulatory cells (Tregs) as a mechanism to suppress #inflammatory responses. The #endocannabinoid system is also involved in #immunoregulation. For example, administration of endocannabinoids or use of inhibitors of enzymes that break down the endocannabinoids, led to immunosuppression and recovery from immune-mediated injury to organs such as the liver. Manipulation of endocannabinoids and/or use of exogenous cannabinoids in vivo can constitute a potent treatment modality against inflammatory disorders. This review will focus on the potential use of cannabinoids as a new class of anti-inflammatory agents against a number of inflammatory and autoimmune diseases that are primarily triggered by activated T cells or other cellular immune components."

    Executive summary

    - Cannabinoids, the active components of #CannabisSativa, and endogenous cannabinoids mediate their effects through activation of specific cannabinoid receptors known as cannabinoid receptor 1 and 2 (CB1 and CB2).

    - The cannabinoid system has been shown both in vivo and in vitro to be involved in regulating the immune system through its immunomodulatory properties.

    - Cannabinoids suppress inflammatory response and subsequently attenuate disease symptoms. This property of cannabinoids is mediated through multiple pathways such as induction of apoptosis in activated immune cells, suppression of cytokines and chemokines at inflammatory sites and upregulation of FoxP3+ regulatory T cells.

    - Cannabinoids have been tested in several experimental models of autoimmune disorders such as #MultipleSclerosis, #RheumatoidArthritis, #colitis and #hepatitis and have been shown to protect the host from the pathogenesis through induction of multiple anti-inflammatory pathways.

    - Cannabinoids may also be beneficial in certain types of #cancers that are triggered by #ChronicInflammation In such instances, cannabinoids can either directly inhibit tumor growth or suppress inflammation and tumor angiogenesis.
