Completed the #DLS safety training once more for next month's visits. Only two answers wrong and again one was to do with a radioactive source which we are unlikely to use in a #MX or #SAXS experiment. The other being the emergency phone number (which should be written on the phones anyway!).

Could the european #synchrotron get a common proposal /sample submission protocol developed for #MX and #SAXS. So much of my time seems to be spend reformatting / re-entering the same information in different formats and then getting the relevant user office complaining it isn't quite right.

The semiannual fight with the
Synchrotron SOLEIL website for submitting an application for more SAXS beamtime.

It was working fine earlier in the week when I downloaded the previous application.

There was a reason I delegated it up (but boss is on holiday this time) 😼

Thrilled to see this very interesting work by Giorgia Greco and colleagues represented as frontispiece for Small Methods! For the first time SAXS/WAXS techniques were used for an in-depth characterization of the AlCl4 anions intercalation process in graphite.

Hinter der SAXS-Probe (bei uns häufig einzelne biologische Zellen, aber bei befreundeten Gruppen auch Mikrokristalle in gleichzeitiger #SAXS-/#WAXS-Geometrie) befindet sich ein #beamstop, um den Primärstrahl (99,9999% der Röntgenphotonen, welche keine Wechselwirkung erfahren haben) abzuschwächen. Denn trotz Unterdruck im Flugrohr würde diese Intensität zu starker Streuung führen und die Ergebnisse verfälschen / die Messung zunichte machen.

Außerdem wäre sonst der #Detektor kaputt …

Das heißt, die Helligkeit unserer Bilder würde ungefähr halbiert; viel schlimmer aber: aufgrund von Streuung würden sie verwaschen/unscharf.

Insbesondere bei Streuexperimenten [nano-#SAXS] muss der Strahl sehr sauber präpariert werden und auch nach der Probe ist Vorsicht geboten. Sehr kleine Proben im µm-Bereich streuen von den einfallenden 10¹² Photonen pro Sekunde vielleicht nur ein paar hunderttausend und eine Million – und diesen paar Quäntchen darf dann nichts „schlimmes mehr passieren“.

The last barrier for #structuralbiology? Giulio Tesei and others in Lindorff-Larsen lab Uni Copenhagen generated ensembles of Intrinsically Disordered Proteins (IDPs) and related them to structural characteristics, such as level of compaction, to annotations such as func. class. But how to relate to what is actually happening in the cell. We're excited to see what imaging techniques such as #cryoem #SAXS super-resolution #microscopy and #nmr might be able to discover!
