    Ce n'est pas du préventif, pas un vaccin, c'est du curatif mais non chimique cette fois et d'une redoutable efficacité. L'autre image connue de la résonance est celle d'une troupe traversant un pont suspendu en marchant au pas cadencé : les pieds frappant le pont tous en même temps le font vibrer, osciller et il peut même casser comme ça. C'est pour ça que les troupes traversant les ponts ne le font jamais en marchant au pas. C'est une image plus précise et plus fidèle que celle d'un pop-corn explosant sous l'effet d'une chaleur qui n'a rien de sonore ou de vibratoire au-delà de l'atome.

    Des chercheurs de l'Université de São Paulo (USP), au Brésil, ont découvert que des ultrasons à haute fréquence, similaires à ceux utilisés lors d'examens médicaux, peuvent éliminer des virus comme le SARS-CoV-2 et le H1N1 sans endommager les cellules humaines. Dans un article publié dans Scientific Reports, ils expliquent comment ce phénomène, appelé résonance acoustique, provoque des modifications de structure des virus jusqu'à ce qu'ils se brisent et soient inactivés.

    "C’est un peu comme abattre le virus en criant. Dans cette étude, on montre que l’énergie des vibrations sonores induit des déformations morphologiques sur les particules virales jusqu’à ce qu'elles en explosent, un phénomène comparable à celui qui se passe avec les popcorns. En déformant la structure du germe, la membrane protectrice du virus appelée enveloppe se déforme et se brise ce qui l'empêche de s’attaquer aux cellules humaines" explique Odemir Martinez Bruno, professeur à l’Institut de physique de São Carlos (IFSC) de l’USP, qui a coordonné l’étude.

    L’inactivation des enveloppes virales avec des ultrasons ouvre de nouvelles perspectives thérapeutiques contre les maladies virales. L’équipe mène d’ailleurs déjà des tests in vitro sur d’autres infections virales telles que la dengue, le Chikungunya et le Zika. Ce traitement alternatif est particulièrement intéressant parce qu'en général le développement des antiviraux est ardu. "Bien que cette approche soit encore loin de l'utilisation clinique, elle constitue une stratégie prometteuse contre les virus à enveloppe parce que le développement d'antiviraux chimiques est complexe et ses résultats sont difficiles à obtenir. En outre c'est une solution écologique parce qu'elle ne génère aucun déchet, n'a aucun impact environnemental et ne favorise aucune résistance virale" explique le Pr Flávio Protásio Veras, professeur à l'Université fédérale d'Alfenas (UNIFAL) et chercheur postdoctoral à la FAPESP.

    Ces recherches ont réuni des scientifiques de différents domaines. Outre les physiciens théoriciens et les acousticiens de l'IFSC, l'initiative a bénéficié de la collaboration de spécialistes du Centre de Recherche en Virologie et du Centre de Recherche sur les Maladies Inflammatoires (CRID), tous deux affiliés à la Faculté de Médecine de Ribeirão Preto (FMRP-USP), à la Faculté des Sciences Pharmaceutiques (FCFRP-USP) et à la Faculté des Sciences et Techniques de l'Université d'État de São Paulo (UNESP). Ces spécialistes ont contribué aux analyses structurales et toxicologiques à l'aide de techniques telles que la microscopie et la réfraction lumineuse. L'initiative a également bénéficié de la collaboration du Pr Charles Rice, professeur à l'Université Rockefeller aux États-Unis et lauréat du prix Nobel de médecine 2020. Le Pr Rice a fourni des virus fluorescents pour visualisations en temps réel.

    C'est de la géométrie

    La découverte a surpris les chercheurs car elle contredit la théorie de la physique classique : la longueur d'onde des ultrasons est bien supérieure à la taille des virus et, en théorie, cette différence de taille empêche toute interaction. "Le phénomène est purement géométrique. Les particules sphériques, comme le sont de nombreux virus à enveloppes, absorbent efficacement l'énergie des ultrasons. C'est cette accumulation d'énergie à l'intérieur du virion qui provoque de façon répétée ces modifications de structure de l'enveloppe virale, ce jusqu'à ce qu'elle se brise. Donc, si ces virus étaient pyramidaux ou carrés ils ne subiraient pas cet "effet popcorn" de résonance acoustique" explique le Pr Martinez-Bruno. Il souligne que l'efficacité de cette technique n'est perturbée en rien par les variants comme ceux observés pendant la pandémie (par exemple Delta puis Omicron) parce que le processus dépend strictement de la forme globale du virus et non de mutations génétiques (ndt : des "récepteurs-harpons" qui tapissent sa surface extérieure et qui déchirent, pour pourvoir entrer dedans, les parois des cellules qu'ils touchent).

    Réglage de la fréquence

    "Cette technique n'est pas destinée à la décontamination. Celle-ci existe déjà. Les ultrasons sont déjà utilisés pour stériliser le matériel dentaire et chirurgical mais ils agissent suivant un phénomène physique différent, appelé cavitation, qui détruit la matière organique" précise Martinez-Bruno. Il explique que la résonance acoustique et la cavitation diffèrent principalement par la fréquence utilisée et leurs effets sur les virus et les cellules. "Alors que la cavitation se produit à basses fréquences et détruit les virus et les tissus par l'implosion de bulles de gaz, la résonance acoustique opère à des fréquences élevées, de 3 à 20 MHz" précise-t-il. Concernant la résonance acoustique, il explique que l'énergie sonore interagit avec la structure virale, provoquant des vibrations internes jusqu'à la rupture mécanique de l'enveloppe virale, ce sans modifier ni la température, ni le pH du milieu. "Il en résulte un mécanisme sélectif et sûr car seul le virus absorbe cette énergie et en est déstructuré, c'est sans risque pour les cellules humaines ajoute-t-il.

Government authorities are still investigating the company’s history, but it appears to have previously operated a lab in Fresno under the name Universal MediTech, where city officials flagged it for investigation regarding improperly stored chemicals.

    "From what is publicly known, the Reedley lab should likely have followed proper biosafety practices to minimize the risks of an #outbreak, and it apparently failed to do so. It could have caused illness, disruption, or even death among local communities and beyond depending on the circumstances of an outbreak. The Centers for Disease Control and Prevention (#CDC) maintains a system of four 'Biosafety Level' standards that are used worldwide for work with dangerous pathogens. Based on the pathogens that were being used at the Reedley lab, it probably should have followed Biosafety Level 3, which involves controlling the airflow inside the lab as well as a host of other practices, equipment, and facility design requirements.

    "Yet, astonishingly, the U.S. government seems to not have even known that the Reedley lab existed until it was discovered by chance by Jesalyn Harper, an observant local city code enforcement officer—the only such officer working full-time in the entire city. Once discovered, the Fresno County and California Departments of Public Health found it to be in violation of local and state codes, including those for registering clinical labs and managing medical waste. Based on our reading of available information, it was likely also in violation of federal Occupational Safety and Health Administration regulations for protecting workers from bloodborne pathogens. But these codes require proactive reporting, and the lab simply never reported any issues to regulators. In slightly different circumstances, it would likely have continued to operate unnoticed for a long time."
