Slightly delayed due to my holidays, but excited to share this new paper that came out online a little while ago in ACS Applied Materials and Interfaces: https://doi.org/10.1021/acsami.4c08428 In it, PhD student Pushpa Rampratap spearheaded our effort to use MAS #NMR to study ECM-like #hydrogels. In a prior publication (https://doi.org/10.1016/j.carbpol.2023.121063) she described methods for preparing 13C enriched version of the polysaccharide hyaluronic acid…. #glycotime #nmrchat #ssnmr

Our latest (review) article is now available online: https://doi.org/10.1042/BST20230731 in the journal Biochemical Transactions. In this invited article “Solid-state nuclear magnetic resonance in the structural study of polyglutamine aggregation”, I have reviewed the contributions that solid-state #NMR #SSNMR has made to our understanding of the mutant protein aggregates in #huntingtonsdisease and related #polyQ disorders. (1/2)

Among many other topics, you can find discussion of how #SSNMR is particularly useful to understand the surface features of protein aggregates (their fuzzy coat), which are often hard to see by cryo-EM and other techniques. (2/2)

Busy week coming - but am definitely looking forward to next week visiting #Cambridge (UK) to discuss #amyloid #ssNMR #ECM #polysaccharides and more. Actually my first visit there, although I spent quite a few years in the 'other' Cambridge (MASS/USA). :-)

Don’t miss the #iupacchains2023 parallel session 10 on Structural Biology and Protein Design at 10:20am - see the talk by my PhD student Raffaella Parlato! She will talk about our work with Wiktor Szymanski’s team (also University of #Groningen) - enabling #photochemical control over #polyQ #amyloid formation & structure. #ssNMR #chemistry

… using in-depth #moleculardynamics simulation to integrate various experimental constraint provides an unprecedented view of the structure of the misfolded protein aggregates, with a central role (of course) for new and existing #ssNMR data. These #polyQ protein fibrils are tough for #cryoEM due to structural and dynamic heterogeneity.

Cool new paper from Joern Schmedt auf der Guenne and coworkers, in the journal #SSNMR: “High resolution solid-state #NMR on the desktop”, presenting a conical MAS rotor design for use in permanent magnets. Looks cool! The team includes Oliver Pecher and Marco Braun from the NMR-service company, which is doing various other interesting things when it comes to #ssNMR hardware! https://www.sciencedirect.com/science/article/pii/S0926204023000346

Well, just got back from the FASEB protein aggregation conference #PACSRC in Ireland. As usual, it was a great event with a never-ending stream of cool science. Quite a few #amyloid structures discusses, based on #ssNMR and #cryoEM . I briefly talked about our own integrative structural biology work on the fibrils made by mutant huntingtin fragments #huntingtonsdisease , as well as our work with aggregation inhibitors (and how they reveal features of the aggregation mechanism) #polyQ

A new preprint from the lab (together with Marthe Walvoort's group) is now on #bioRxiv : https://www.biorxiv.org/content/10.1101/2023.03.16.532902v1 This manuscript presents nice work by Pushpa Rampratap, who did a great job figuring out how to make 13C-enriched hyaluronic acid #polysaccharide for use in #ssNMR . More forthcoming on the applications, but this paper introduces the protocols for producing the high-molecular weight HA. Enjoy! #glycotime #nmrchat #TeamMassSpec (Feedback welcome!)
