On Wednesday I made a small donation to Processing.org, because I'm really liking p5.js for simple graphics work, and because they asked for money.

On Thursday, I discovered q5.js, a fork/reimplementation that appears to be much better designed.

I'm happy to report that I've seen radically improved efficiency of the excellent NEB #Q5 DNA #polymerase for DNA amplification when using the following home-made reagent :-)

Extra-magic PCR buffer (10X recipe):
• 0.5M TrisHCl pH 8.9
• 0.1M Ammonium Sulphate
• 0.3M KCl
• 20 mM MgCl2
• 0.1% Tween 20

This is of course, anecdotal evidence based on a few pairs of long primers and using #genomic DNA as template. The cloned PCR fragments were clean of mutations. #diy

2024 Porsche Macan
While the Taycan was at the dealer for some work, they very kindly leant me a brand new (less than 500 miles on the clock) 2024 Macan. Not, of course, the upcoming EV version of the Macan, but a regular ICE version.

This captures a few of my thoughts on this vehicle, not compared to the Taycan, but to a 2018 Audi Q5 - its Audi badged cousin, but from 6 years ago.
