The humans must return the severed head of the forest god because humans severed it in the first place. But it's interesting that the humans who do that are liminal characters from the fringes of human society - Ashitaka is an indigenous Emishi and San was literally raised by wolves, neither is aligned with the humans who caused the ecological destruction in the first place.
