#FYI #rapamycin #alzheimers #pharmacokinetics
"Rapamycin, an mTOR inhibitor used clinically for immunosuppression, shows promise for repurposing in age-related disorders including Alzheimer's disease (AD).
This open-label pilot study investigated the pharmacokinetic properties of weekly oral rapamycin in [...] early-stage AD.
Findings [...] support fixed-dose regimens in future trials."https://biologists.social/@medrxivpreprint/114188319062962075

i am using clobetasol sparingly (ok once a day) but it is helping me creep back to semi normalcy? I have used a bevy of other topical steroids and you have to be really careful not to go over the line - one minute it is helping and next minute it is hell...there are a plethora of monoclonal Ab peptides on the way with v good 3rd stage trials results; no miracle cures yet for psoriasis, i think they are still drilling down a bit and all various interleukins/pathways #il23 #pharmacokinetics

A unifying #model to explain frequent #SARS-CoV-2 #rebound after #nirmatrelvir #treatment and limited prophylactic #efficacy https://www.nature.com/articles/s41467-024-49458-9
Here authors develop a mathematical model for viral-immune dynamics & nirmatrelvir #pharmacokinetics and suggest that early treatment preserves susceptible cells & blunts innate immune response without fully eliminating infection. They project that treatment extension for 10 days may overcome this effect.

What this boils down to is using mathematical/statistical models to help explain and predict what the body does to the drug (#Pharmacokinetics, #PK) and what the drug does to the body (#Pharmacodynamics, #PD) - these are often combined to produce #PKPD or #ExposureResponse models.

Pharmacometrics rolls up modeling and simulation for #Pharmacokinetics, #Pharmacodynamics, and #DiseaseProgression, with a focus on populations and variability. A major focus is to understand variability in drug response, which can be predictable (e.g. due to differences in body weight or kidney function) or unpredictable (differences between subjects seem random, but likely reflect a lack of knowledge or data).
