I am working on my first #Minion metabarcoding run. I've got a library of dual-indexed amplicons (ITS2). Ran it through the Minion, base called, and now I'm working on demultiplexing.
I have discovered that some of my reads contain multiple amplicons. i.e., a single line in the fastq file includes index1-primer-read1-primer-index2-index3-primer-read2-primer-index4
Is that a problem with my library, the sequencing process, or is it something I can fix at this stage (i.e., computationally)?

Respect to whoever went the extra mile to make this Minion letterbox.
I stopped to photograph it at the entrance to a rural driveway in Lyttelton Harbour. I like how yellow reflectors have been added to it can be seen by car traffic.
Some skilled parent must be very popular with their children. 😀

@nanopore DRS sequencing temperature was lowered to 30/31C (#P2 / #MinION) in #minknow v24.06 (and all recent versions) slowing down RNA helicase from 130 bps to 100-110 bps, lowering yield per hour and affecting signal readouts.
In effect, recent and older #RNA004 runs differ in base- and modification calling quality, possibly pA tail estimates.
This change was not widely announced to the community apart from a brief mention in software release notes!

I have accepted the fact that that I don't have profiles on #spotify that it is broken as hell, but now that I see my #letterboxd also has "most watched actors" filled with people I did not know were the voices of the #Minion movies, I really need to push them to add a feature that makes it easy to separate "watched with kid" vs "watched with grown ups" 😅🤣
