In today’s #positivenews it’s been confirmed that the heating system I was repairing for the old guy who lives up the road is fully functioning. Ugh, someone botched the #WikiPedia article on #IVT 's. It's prefixed with:

Help! I have a problem with #IVT in vitro transcription:
I created a set of plasmids with T7 promoter & poly-A tail. my gene (~4kb) is inserted in between. I have several variations of the same gene. In all cases, I get 2 products: the correct one and a shorter (~2.8kb) one.
Any idea why? How do I solve it?I use the HiScribe T7 High Yield RNA Synthesis Kit (NEB) with 3"-O-Me-m7G(5")ppp(5")G RNA Cap Structure Analog.
IVT for GFP works well with just one product of correct size. -
#MathMonday I love it when there are useful, “everyday”, applications of a mathematical theorem. Here’s one of my favorites: Take a level table wobbling on uneven ground. The Intermediate Value Theorem guarantees that rotating the table in a circle, less than a quarter turn (for a 4-legged table), will find a spot where the table sits perfectly without wobbling! No need to shove napkins under the “short” leg. (#Proof available upon request.) #calculus #IVT
