Well, we've got an NHS blood test booked next Friday morning, which will include hormone level tests for our gender clinic, so sadly we need our estradiol level to be between 400 and 600 pmol/L so that they don't complain at us :Sighing_Face:

Our estradiol trough goes up by roughly 125 pmol/L for each 1 mg we inject, so this should put our estradiol level somewhere between about 500 and 600 pmol/L.

If enough folks asks for it, we'll do a poll to guestimate what our estradiol test result will be after the test 🩷

Delayed again by a few hours today. Didn't think we'd have the spoons to do it at all tonight, but had a slight rally. Weird injection though. Not painful, just uncomfortable when pulling out the needle. No blood spurt or anything. Might have been we loosened our grip on the z-track or something, or because we did more walking today by far than we have typically done for 6-12 months. Anyway, all went in fine and no pain afterwards.

FYI, for anyone who hasn't seen our earlier posts and thinks something like, "Golly! That's a high dose!", it's actually not 😅

Whilst simulations typically suggest this would give us a high estradiol level, our trough only goes up by about 125 pmol/L for each 1 mg of estradiol enanthate injected.

As such, we're looking at a trough of about 875 to 938 pmol/L, and a peak under 1100 pmol/L, which is a perfectly-safe range for estradiol injections where a higher-level is desired for mood and for applying negative pressure to the HPG axis :TransHeart:

Should have done it yesterday evening from about 18:00 onwards, so it's about 18 hours later than usual 😅

Though we often do our injection a little later than planned, this is the first time in a while that we've actually neglected to do it until the next day 🤦‍♀️

In fairness to ourselves though, we were very tired yesterday (and still are today) and had a lot of other things on our mind :catgirl_sleepy:

Plus we're using estradiol enanthate, which has a longer elimination half-life than both estradiol cypionate and estradiol valerate, so any level drop would have been minimal.

Our eventual aim will be to switch over to a monthly injection of estradiol undecylate (at 100 mg/mL), but we'd prefer to keep a more-stable, higher level until our gonads get yeeted and become nonads 🤭

(In case anyone's wondering, we added clarification to give a slightly range on the dose today and last week due to tiredness, as that might have made our dose less accurate.)

Did it more-or-less on time, but wasn't as easy a one today. Considering the relative lack of sleep we've had recently (especially today even with a nap), that probably contributed.

Still, even a less-easy auto-injector injection is noticeably much easier than a difficult manual injection for us. It might not have been flawless, but we're still talking in terms of "slightly felt it, but not actually painful". More like "minor discomfort" at worst.

Left it late again today because other stuff needed doing first, rather than out of active choice.

Had a small bleed (because of course we did right now 🤦‍♀️) but caught it immediately because we're just expecting small things like that to happen right now and staying on guard accordingly. Otherwise painless and easy :TransHeart:

For anyone looking at 7.5 mg on an E2-level simulator and getting confused: all the online simulators massively overestimate E2 level for us. We've adjusted ours, based on several blood tests.

For each 1 mg we inject, our trough (blood drawn in morning; injection in evening that same day) goes up by about 125 pmol/L. Allowing for a margin of error, we like to keep our trough >= 900 pmol/L.

If you use the Estrannaise simulator, our results with 7.5 mg weekly are what it suggests someone would get with 4 mg. (It overestimates E2 level for us by about 88%.)

Just mentioning this, as folks sometimes forget that simulators are just simulations and won't tell you 100% what E2 level you'll get from a dose.

Hey folks

A friend of ours has been experiencing something odd that we've not seen before:

• Her estradiol (E2) pmol/L per mg level has been gradually rising over the last 305 days.

... and we lack the understanding of statistical analysis to be able to analyse it properly 😞

All injections were done weekly, using estradiol enanthate compounded at 40 mg/mL from the same homebrewer.

We don't think markdown tables work in Glitch-SOC, so here's all the data we have in a non-tabular form.

We're not sure how to use the data to model how to get her pmol/L per mg stable, rather than steadily increasing.

The desired aim is to keep her pmol/L trough roughly between 750 to 900 pmol/L, without it increasing steadily over time.

We think this might mean moving to a 8-10 day injection cycle and a lower dose (0.10 to 0.12 mL; 4 to 4.8 mg), but we are not sure how to do it.
