    Abstract

Background: Usutu virus (Orthoflavivirus usutuense, USUV), a neurotropic arthropod-borne RNA virus of the family Flaviviridae, is a zoonotic virus that has spread throughout the European continent over the last three decades, since its emergence in Italy in 1996. However, no cases of USUV have been reported in Portugal so far.

Material and methods: In the scope of an active surveillance program for Orthoflavivirus, we collected growing feather samples from 249 red-legged partridges (Alectoris rufa) hunted in southern Portugal during the 2021-2023 hunting seasons. Samples positive for USUV were subjected to whole genome sequencing and strain characterization.

Results: Two partridges tested positive for USUV. Phylogenetic analyses of whole and partial genomes assigned the USUV strains to the African 3 lineage, specifically the African 3.1 sub-lineage.

Conclusions: Our study confirms, for the first time, the circulation of USUV in wild birds in Portugal. Active surveillance of hunted partridges proved to be a useful, accessible, and cost-effective method for USUV monitoring, further supporting their value as effective sentinels for Orthoflavivirus surveillance. Given the ongoing circulation of USUV and the increasing risk of its spillover to other domestic and wild animals, and humans, additional efforts are needed to improve virus surveillance in Portugal from a One Health perspective.

  8. The above ☝️ study from Slovakia had to contend with three different geographically co-circulating flaviviruses: West Nile Virus, Usutu Virus, and Tick-Borne Encephalitis #Virus.

For comparison, here's a study from the Amazon, looking at cross-reactivity among three different #flavivirus: Dengue Virus, Yellow Fever Virus, and Zika Virus.

  9. Have you ever had a weak positive test for West Nile #virus only to test negative on a subsequent test?

One recurring problem with #epidemiology of #flavivirus #InfectiousDisease is that antibody tests for one flavivirus happen to be highly crossreactive with other flaviviruses.

That is a problem addressed in:

  10. Ilheus #virus is a poorly known mosquito-transmitted neurotropic #flavivirus that can cause serious brain infections.

Like some other neurotropic viruses, it is no longer detectable in blood once the symptoms have begun.

A new meta-analysis aimed to quantify how common it is and found that in Brazil approximately 8% of the population showed serological exposure.

Now, many RNA viruses have a viral protein called a #viroporin, which creates pores in membranes such as the endoplasmic reticulum.

In some viruses, such as an #enterovirus, the viroporin is well-known. (2B protein)

In other viruses, the viroporin is better known for something else, as the viroporin function is not well appreciated.

For example, in a #flavivirus, the NS2A protein is known as a capsid-assembly protein. Its function as a viroporin is less well-known.doi.org/10.1080/21505594.2017.
