    Our eFIB-SEM (as per Xu et al. 2017 elifesciences.org/articles/259 ) runs 24/7 for up to 8 weeks, when it runs out of source. And it's essential that it does so, uninterrupted, since it's imaging at nanometre resolution, milling 2 nm slices at a time with an ion beam and then repeatedly imaging the block face. The process is unsurprisingly very sensitive; was hard enough to overcome the reheat cycle of the instrument itself (every 60 hours or so) by detecting it, de-engaging the beams and then seamlessly re-engaging with the block face.

    The MS Windows computer deciding to reboot on its own is unacceptable.

  22. Remembering George Palade, born #OTD in 1912. His use of cell fractionation and electron microscopy led to many discoveries and laid a foundation for modern cell biology.   23. New preprint from Benoit Zuber's group showing that FerriTagging (a method for sticking a ferritin particle on a protein of interest so that you can see it) is compatible with cryo-electron tomography.

    This is a scanning electron microscopy image of the cranial sutures in the rat skull. 🔬💀🐀 These are fibrous joints where the bones of the skull interdigitate, allowing for flexibility and growth during infancy and early childhood before fusing in adulthood.
