On Saturday, April 25 we celebrate our troupe's big 20th birthday so we're doing a retrospective countdown!

Year 6 (2011 - 2012) was when we officially were on the schedule for Pennsic for the first time with an encore of "The Horoscope"!

Year 7 (2012 - 2013) saw some of our biggest performances ever as we celebrated the 50th anniversary of "Doctor Who" with several parodies including our Defenders of Neo-Nippon 3 Team Henshin-H anime crossover "Dottore Who & the Curse of the Fatal Fanservice"!

Year 8 (2013 - 2014) when we played on the Pennsic Amphitheatre stage for the first time with a double feature of commedia dell'arte adaptations of two farces by the Roman poet Plautus: "Miles Gloriosus" and "Aulularia: the Pot of Gold"!

Year 9 (2014 - 2015) when we prepared for trouble and made it double with our Defenders of Neo-Nippon 3 Team Henshin-H anime parody "QB's Used Wish Emporium" at at our alma mater Case Western Reserve University's Anime Marathon!

Year 10 (2015 - 2016) we went to a Patchwork Multiverse that's right over there with a bawdy commedia dell'arte parody of Star Wars and one of our punniest titles in "Looking for Love in Alderaan Places" as our first full length performance at Cleveland ConCoction!

Year 1 (2006 - 2007) saw "The Days After the Days of the Valentine's" as our first performance under the banner of The Confused Greenies, a short modernized commedia dell'arte farce for the combined Carnevale & Mardi Gras celebration at our alma mater of Case Western Reserve University!

Year 2 (2007 - 2008) we adapted "The Horoscope" at CWRU as our first traditional commedia dell'arte full three-act play which we'd later encore several times as one of our faves!

Year 3 (2008 - 2009) we expanded from our traditional commedia dell'arte roots to study a different culture's historic improvised theatre in our first "Mad Words: the Art of Kyōgen" shows at CWRU's Anime Marathon!

Year 4 (2009 - 2010) the troupe got really active including modernized commedia dell'rte of both "Doctor Who" and our first musical parody of "The Rocky Horror Picture Show"!

Year 5 (2010 - 2011) we kept the momentum going with an even bigger season, the highlight the premiere of "Dottore Who & the Zealous Ones" where our bumbling parody time traveler meets the most famous commedia dell'arte troupe in history during a foolish alien invasion of the Italian Renaissance! Still one of our finest performances to date that also introduced our signature short play "It All Makes Sense Now!"

In two months we celebrate our 20th birthday - but earlier this week (the same day Carnevale ended and the new lunarsolar year began) marked the 19th anniversary of our very first performance under the banner of The Confused Greenies and i Verdi Confusi! Appropriate for the occasion, that was also staged to entertain Carnevale at our alma mater Case Western Reserve University! And over 450 performances since then!

It's a big sportsball weekend with not only the Superbowl tomorrow but also the Winter Olympics in northern Italy! Perhaps later this year, maybe we'll encore our commedia dell'arte about a Renaissance Italy predecessor to football, "A Comedic Farce of a Fraudulent Cow in Calcio Fiorentino"!

Happy National Take the Stairs Day if you can (and okay if you can't)! We're taking steps toward our 20th anniversary and reminded of one of our earliest shows when we learned the Secrets of Cartoon Physics at Case Western Reserve University that in a farcical comedy any object (or foolish harlequin) falling from an extreme height will be slower than someone frantically running down the stairs of every story (often with repeated clips to save budget). So remember that if you're ever in a 'toon 'verse!

Myszom „wróciła pamięć”. Naukowcy cofnęli objawy Alzheimera w laboratorium, ale ostrzegają przed hurraoptymizmem

To brzmi jak scenariusz filmu science-fiction: naukowcy podali myszom z zaawansowaną demencją eksperymentalny związek chemiczny, a te odzyskały zdolności poznawcze.

Wyniki badań opublikowane w „Cell Reports Medicine” dają nadzieję, ale droga do aptek jest jeszcze bardzo długa i wyboista.

Zespół amerykańskich badaczy pod kierownictwem prof. Andrew A. Piepera z Case Western Reserve University ogłosił sukces w badaniach przedklinicznych nad chorobą Alzheimera. Kluczem do sukcesu okazał się związek o nazwie P7C3-A20.

Co dokładnie się stało?

Naukowcy pracowali na myszach genetycznie zmodyfikowanych tak, by rozwijały ludzką wersję choroby Alzheimera (zarówno poprzez mutacje białka amyloidu, jak i białka tau – dwóch głównych winowajców choroby). Eksperyment podzielono na dwa etapy:

• Prewencja: podano lek młodym myszom (2-miesięcznym). Związek P7C3-A20 skutecznie zapobiegł rozwojowi choroby.
• Leczenie (tu uzyskano przełom): lek podano myszom starszym (6-miesięcznym), u których choroba była już zaawansowana. Wynik? Gryzonie odzyskały zdolności poznawcze, a procesy degeneracyjne w mózgu zostały zatrzymane lub cofnięte.

Jak to działa? Kluczem jest energia mózgu

Badacze odkryli, że w chorobie Alzheimera kluczowym problemem jest spadek poziomu koenzymu NAD+. Jest to cząsteczka niezbędna do metabolizmu komórkowego – mówiąc prościej: to paliwo dla komórek. Gdy poziom NAD+ spada, komórki mózgowe „głodują”, pojawia się stan zapalny i przestają one radzić sobie z usuwaniem toksycznych białek.

Związek P7C3-A20 działa jak regenerator – przywraca zdolność mózgu do utrzymania właściwego poziomu NAD+, co pozwala komórkom „naprawić się” i wrócić do pracy. Co ważne, robi to bezpieczniej niż dostępne na rynku suplementy (prekursory NAD+), które przy nieostrożnym stosowaniu mogą podnosić poziom tego koenzymu tak bardzo, że staje się to toksyczne lub wręcz rakotwórcze.

Dlaczego nie otwieramy szampana?

Mimo że prof. Pieper mówi o „przesłaniu nadziei”, a inni eksperci (jak cytowana przez BBC prof. Tara Spires-Jones) przewidują przełom w ciągu 5-10 lat, musimy zachować chłodną głowę.

Przede wszystkim mysz to nie człowiek. Nasze mózgi są znacznie bardziej skomplikowane. Historia medycyny zna tysiące leków na Alzheimera, które świetnie działały w klatkach laboratoryjnych, a kompletnie zawodziły w testach klinicznych na ludziach.

To nie jest lek z apteki: P7C3-A20 to na razie związek chemiczny do badań. Choć w internecie już pojawiają się strony oferujące sprzedaż „cudownych fiolek” z tą substancją, naukowcy i lekarze stanowczo odradzają kupowanie ich na własną rękę. To niesprawdzone, nieprzetestowane na ludziach substancje, które mogą być śmiertelnie niebezpieczne.

Na „lek na Alzheimera” musimy poczekać do wyników badań klinicznych na ludziach. Na razie mamy dowód, że uszkodzenia mózgu – przynajmniej u ssaków – nie muszą być nieodwracalne.

This week, 14 years ago, we staged our first anime parody play... which included K-Pop dancers... hunting demons.... Huh...

For the Fall 2011 Case Con Anime Marathon at Case Western Reserve University we not only wrote a parody mashing together the anime being watched that semester (including "Steins;Gate" and "Higurashi") but we also invited the local Niji Dancers to be an integral part of the plot where their K-Pop dance moves were combined with time-travel to track down and defeat a kawaii cyber-demon from Venus! . . . . It... was a thing...

In all the focus on Commedia Day, we forgot to mention that this month marks the TWENTIETH anniversary of the first commedia dell'arte performance by our Artistic Director and Capopcomico Jess Rudolph while a student at Case Western Reserve University! That foundational show (for La Dolce Vita's Carnevale) helped pave the way for the formation of The Confused Greenies of Players' Patchwork Theatre Company a little over a year later!
